Hello
So we have had ICSI a few times and had no fertilisation and although they don't know how to improve our chances in the lab, they want my hubby in today to give a sample for DNA Frag and aneuploidy screening.
I'm quite happy to go along with it because, well, why not, but i'm not exactly sure what significance the results will have.
Will the DNA Frag be a percentage and the Aneuploidy simply be normal or abnormal?
If the DNA Frag come back a high percentage what does it mean for us?
If the aneuploidy comes back abnormal what does it mean for us?
I suppose i'm wondering whether they are just hoping to convince us that we should just use donor sperm and stop bringing their stats down and making them look clueless  |

The DNA Fragmentation Index results will come back as a % of sperms cells containing measurable DNA damage -
'< 15% DFI = excellent fertility potential
> 15 to < 30% DFI = good fertility potential
> 30% DFI = fair to poor fertility potential*
The statistically significant DFI threshold for subfertility has been established at > 30%.
Normal full-term pregnancies are possible with an elevated DFI, but the higher the level of fragmentation, the greater the incidence of reduced term pregnancies and miscarriage.'

The Sperm Aneuploidy results will show the Chromosomes Disomy (Gonosomal and Autosomal) as a % and the total Aneuploidy as a % too. If these figures are higher than the normal values, then they indicate issues, as -
'Studies have shown that sperm with a high rate of aneuploidy have a negative impact on pregnancy rate and are associated with recurrent pregnancy loss.'
Like I have said before ... the test results do not definitively mean that the sperm is unable to produce a healthy baby, just an indication of the probability, IYKWIM.

Hiya,
Yes thanks Maple i had checked the links
I suppose that they just still left me a little confused why the clinic thinks that there is a good reason for us having them when they seem to suggest that it's just more diagnostics for the part after we achieve fertilisation, when we can't achieve fertilisation.
I was wondering why other people were having the tests in case they had something that related more to my situation xx |

Hi Louise,
I guess your clinic does not know what is the cause of your fertilisation issues, and these tests are the only more thorough sperm tests available to them (whether it is truly applicable in your case or not) ... but it is really all they have got and it the tests come back really bad, they can use it as part of the reason, IYKWIM.
Besides testing your eggs with PGD/aCGH, they really have no other diagnostic tests available to help them/you to find the root cause.

Yeah, i suspected as much, it's a bit frustrating.
Why haven't you managed to be successful yet? Have you exhausted all diagnositics too? |

Hi Louise,
Yes, I have more or less exhausted everything diagnostic. I have tried DE and immunology, but still had multiple miscarriages ... my post myomectomy (possibly scarred) uterus may be at fault or the sperm may be at fault. No expert can tell me what is causing the problem.
My DH and I are not in a position to use DS ... so far, the sperm tests had not come back extremely bad (but not great either).
Hope you have a less frustrating and tortuous journey ... I really feel for you. But keep fighting ... with your determination, you will get there.
